INTRODUCTION TO SESSION

ON

"GENETIC THERAPY OF HUNTINGTON’S DISEASE"

Huntington’s Disease (HD) is an inherited autosomal dominant, neurodegenerative disorder characterized by a constellation of motor, cognitive, and psychiatric features. HD is caused by an abnormally expanded, unstable trinucleotide (CAG) repeat in one allele of the huntingtin (HTT) gene. The mutant HTT gene is expressed as a long polyglutamine tract in the mutant protein (mHTT) that confers a toxic gain of function that results in progressive neuronal degeneration in the CNS.

Up until the discovery of the HD gene, therapy for HD was limited to agents that alleviate symptoms, because no treatment had been shown to prevent onset or to slow progression. However, new disease modifying approaches that target HD gene expression show great promise.

In this session, we will review some of the approaches that target the HD gene. Improvements in the delivery and distribution of such agents as well as the development of objective biomarks of disease and of HTT lowering outcomes have brought these potential therapies to the forefront of HD research, with clinical trials in patients already underway.
